Summary

The project \'UbiArchitect\' set out to further understand architecture in ubqiutiin chains, and to reveal and understand new ubiquitin signals, using a variety of biochemical, mass-spectrometric, biophysical and structural biology techniques.

the ubiquitin system presents a large and mostly untapped translational resource, but the complexity of ubiquitin signals has been daunting for pharmaceutical efforts.

the overall objectives are to develop new methods and techniques to study ubiquitin signals by mass-spectrometry, and apply them to medically relevant pathways. a particular focus is on Parkinson;s disease.

Work performed

The 5 year project was terminated after year 1 as the principal investigator moved to Australia.

During the first year, the group addressed AIM 2a, mechanistic understanding of PINK1/Parkin mediated mitophagy, providing two Nature papers revealing the first structure of PINK1 in complex withubiquitin and a nano body, and the structure of the active form of Parkin. Significant progress was also made on AIM 1, which will be published in 2019.

Final results

the project was terminated, and will be continued in Australia. We expect to address all aims in the next few years.
